deoxy Amidites and Supports, labile protection

ChemGenes offer various DNA phosphoramidites with labile base protecting groups. Even though, DNA deprotection is relatively simple compare to RNA, proper care needs to taken while choosing the correct conditions and deprotection times.

Cleavage of Oligo from the support and deprotection of protecting groups can be achieved by using any of the following deprotection conditions. (Deprotection time ranges are dependent on the oligo chain length)

  • Conc. aq. Ammonia (30%) at rt for 4 hours (for shorter sequences ~20 to 30-mer).
  • Conc. aq. Ammonia (30%) at 37°C for 12-16 hours (this condition is recommended for the longer oligonucleotides of ~40-mer or more).
