5-EthylThio Tetrazole has been demonstrated to be a superior coupling reagent for oligo ribonucleotide synthesis.

This reagent increases the stepwise coupling efficiency by


2-3% compared to the conventional activator, 1-H Tetrazole (B.Sproat, F. Colonna, B. Mullah, D. Tsou, A. Hampel, R. Vinayak, Nucleosides & Nucleosides, 14,255,1995).

The total oligonucleotide yield and quality have been shown to increase dramatically when using 5-EthylThio Tetrazole. Perhaps the greater acidity and stability of 5-EthylThio Tetrazole account for its superior performance.

This reagent is especially suitable for large scale oligo ribonucleotide synthesis, and also very suitable for DNA synthesis.

Advantages in DNA/RNA Synthesis:
• Higher reactivity than 1-H Tetrazole
• Better solubility than 1-H Tetrazole
• Avoid crystallization in synthesizer
• Less expensive than 1-H Tetrazole
• Coupling efficiency increased 2-3% during oligonucleotide synthesis
