1. PAC-Protected nucleosides phosphoramidites must be used for efficient base deprotection using ammonia free oligo
deprotection solution.

2. Oxalyl CPGs must be used for ammonia free oligo deprotection solution to cleave the oligo from the support. The standard succinyl CPGs inadequately deprotect when using ammonia free oligo deprotection solution. The CPG (1 ΅mole scale synthesis) after oligo synthesis is washed with ether, air dried, and is placed in a vial with a septum cap. The 1 ml of the ammonia free oligo deprotection solution is added using a syringe.
